From Amritsar(Punjab) to Manali(Himachal Pradesh) road way is approx 400 km on NH-20 via (Batala--Gurdaspur--Pathankpt--Bungal--Dharamshal-Mandi-Bhuntar-Manali).
There is another way to reach at Manali via Hoshiarpur---Manadi---Bhuntar--Manali on NH-70.

Volvo buses reach late night at Amritsar-Chandigarh, so I advice you to pick Volvo from Delhi.

Amritsar to Nankana Sahib from International Bus Terminal. Youth Hostel, G.T. Road Amritsar at 9:30 A.M.(IST).
From there you get a cab or taxi to reach at Manali.
And
Every Tuesday and Saturday goes Punjab roadways bus to Amritsar:
Nankana Sahib to Amritsar from Nankana Sahib at 9 A.M.(IST)

That is better idea to travel by cab/taxi to Manali.
The total distance of Amritsar to Manali is 400kms approx and it should take about 8-10 hours. Taxi/Cab charges would be depend on type of cab hired (4 seater /10 seater /12 seater). Roughly the charges should vary from 9/- per kms to 15/- per kms.

You got to 2 ways for travel to Manali from Amritsar.
Amritsar to Pathankot then on Nurpur road for Mandi via Baijnath, Jogindernagar. From Mandi take the Kullu road to Manali. Approx distance 390 km and the other way is from Amritsar for Jallunder then to Hoshiarpur via Adampur and from there to Mandi via Una. Mandi onwards its the same route via Kullu to Manali as in the first option. This route covers lot of plains so you will be able to drive faster. It will approx 375km full route to Manali. The first option hills start from Pathankot. You will be covering less distance also roads are not good nearby Nurpur and near Jogindernagar also roads were not very good but you still doable on decent speed. The scenery on this route is better though.
